| Tunicamycin | N-linked glycosylation is a highly regulated post-translational modification that is involved in protein folding and conformation, oligomerization, sorting, cell-cell interactions, and targeting of proteins to sub- or extra-cellular locations. It is initiated in the endoplastic reticulum with the transfer of a carbohydrate moiety to an asparagine residue within a specific amino acid consensus sequence and then further processed in the Golgi whereupon a mature glycoprotein is exported through the secretory machinery to the plasma membrane. Tunicamycin, a mixture of nucleoside antibiotics, is a specific inhibitor of N-linked glycoslylation that blocks the first step of glycoprotein synthesis thereby inducing protein unfolding. At 500 nM, it can impair the function of several receptor tyrosine kinases, including EGFR, ErbB2, ErbB3, and IGF-IR.1 Also at 500 nM, it radiosensitizes U251 glioma and BXPC3 pancreatic adenocarcinoma cells to chemotherapy.1 Tunicamycin impairs ALK phosphorylation Group: Biochemicals. | Vancomycin Hydrochloride | Vancomycin is an aminoglycoside antibiotic, used in the treatment of infections caused by Gram-positive bacteria. vancomycin is not active against Gram-negative bacteria (except some non-gonococcal species of Neisseria). The main target of this antibiotic is the D-alanyl-D-alanine terminal dipeptide of peptidoglycan precursors, used by bacteria for constructing their cell walls. This prevents the reaction used to link peptidoglycan precursors together from taking place. Vancomycin binds with the substrate, not the enzyme, in contrast to the way penicillin inhibits peptidoglycan synthesis.The large hydrophilic molecule is able to form hydrogen bond interactions with the terminal D-alanyl-D-alanine moieties of the NAM/NAG-peptides. Under normal circumstances, this is a five-point interaction. This binding of vancomycin to the D-Ala-D-Ala prevents cell wall synthesis in two ways. It prevents the synthesis of the long polymers of N-acetylmuramic acid (NAM) and N-acetylglucosamine (NAG) that form the backbone strands of the bacterial cell wall, and it prevents the backbone polymers that do manage to form from cross-linking with each other. | Verruculogen | Verruculogen is a tremorgenic mycotoxin first isolated from Penicillium verruculosum in 1972 and the structure resolved as an indole alkaloid in 1974. Subsequent investigations have shown that verruculogen is produced by several species of Penicillium and Aspergillus and its presence is a useful taxonomic phenotypic marker. The tremorgenic action of verruculogen is complex but is associated with increases in spontaneous glutamate and aspartate release, decreases in GABA levels and, at toxic doses, an increase in the number and decrease in the affinity of DHP receptors in rat cortex. In in vitro guinea pig ileum preparations, verruculogen causes an increase in contractile responses due to electrical field stimulation, attributed to enhancement of acetylcholine from presynaptic nerve terminals.
