(6-4)DNA photolyase A flavoprotein (FAD). The overall repair reaction consists of two distinct steps, one of which is light-independent and the other one light-dependent. In the initial light-independent step, a 6-iminium ion is thought to be generated via proton transfer induced by two histidines highly conserved among the (6-4) photolyases. This intermediate spontaneously rearranges to form an oxetane intermediate by intramolecular nucleophilic attack. In the subsequent light-driven reaction, one electron is believed to be transferred from the fully reduced FAD cofactor (FADH-) to the oxetane intermediate thus forming a neutral FADH radical and an anionic oxetane radical, which spontaneously fractures. The excess electron is then back-transferred to the flavin radical restoring the fully reduced flavin cofactor and a pair of pyrimidine bases. Group: Enzymes. Synonyms: DNA photolyase; H64PRH; NF-10; phr (6-4); PL-(6-4); OtCPF1; (6-4) PHR; At64PHR. Enzyme Commission Number: EC 4.1.99.13. CAS No. 37290-70-3. Storage: Store it at +4 ?C for short term. 6G-fructosyltransferase Inulins are polysaccharides consisting of linear or branched D-fructofuranosyl chains attached to the fructosyl residue of sucrose by a β(2?1) linkage. This enzyme catalyses the transfer of the terminal (2?1)-linked -D-fructosyl group of an inulin chain onto O-6 position of the glucose residue of another inulin molecule. For example, if 1-kestose [1F-(β-D-fructofuranosyl)sucrose] is both the donor and recipient in the reaction shown above, i.e., if m = 1 and n = 1, then the products will be sucrose and 6G-di-β-D-fructofuranosylsucrose. In this notation, the superscripts F and G are used to specify whether the fructose or glucose residue of the sucrose carries the s...nd n ≥ 0 for the acceptor. Group: Enzymes. 7,8-didemethyl-8-hydroxy-5-deazariboflavin synthase Binds a [4Fe-4S] cluster. The cluster is coordinated by 3 cysteines and an exchangeable AdoMet molecule. The first stage of catalysis is reduction of the 2 AdoMet to produce 2 methionine and 2 5'-deoxyadenosin-5-yl radicals that extract a hydrogen from each of the substrates permitting the condensation of the two. The overall reaction catalysed is the transfer of the hydroxybenzyl group from 4-hydroxyphenylpyruvate (HPP) to 5-amino-6-ribitylaminopyrimidine-2,4(1H,3H)-dione to form 7,8-didemethyl-8-hydroxy-5-deazariboflavin (FO). 7,8-Didemethyl-8-hydroxy-5-deazariboflavin is the chromophore of the hydride carrier coenzyme F420.
