Besides interconverting citrate and cis-aconitate, it also interconverts cis-aconitate with isocitrate and, hence, interconverts citrate and isocitrate. The equilibrium mixture is 91% citrate, 6% isocitrate and 3% aconitate. cis-Aconitate is used to designate the isomer (Z)-prop-1-ene-1,2,3-tricarboxylate. homoaconitatehydratase
Requires a [4Fe-4S] cluster for activity. The enzyme from the hyperthermophilic eubacterium Thermus thermophilus can catalyse the reaction shown above but cannot catalyse the previously described reaction, i.e. formation of (R)-homocitrate by hydration of cis-homoaconitate. The enzyme responsible for the conversion of cis-homoaconitate into (R)-homocitrate in T. thermophilus is unknown at present but the reaction can be catalysed in vitro using aconitatehydratase from pig (EC 4.2.1.3). Group: Enzymes. Synonyms: homoaconitase; cis-homoaconitase; HACN; Lys4; LysF; 2-hydroxybutane-1,2,4-tricarboxylate hydro-lyase (incorrect). Enzyme Commission Number: EC 4.2.1.36. CAS No. 9030-68-6. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder. EXWM-5027; homoaconitatehydratase; EC 4.2.1.36; 9030-68-6; homoaconitase; cis-homoaconitase; HACN; Lys4; LysF; 2-hydroxybutane-1,2,4-tricarboxylate hydro-lyase (incorrect). Cat No: EXWM-5027.
2-methylcitrate dehydratase
The enzyme is specific for (2S,3S)-methylcitrate, showing no activity with (2R,3S)-methylcitrate. The enzyme can also use cis-aconitate as a substrate but more slowly. Both this enzyme and EC 4.2.1.3, aconitatehydratase, are required to complete the isomerization of (2S,3S)-methylcitrate to (2R,3S)-2-methylisocitrate. Group: Enzymes. Synonyms: 2-methylcitrate hydro-lyase; PrpD; 2-hydroxybutane-1,2,3-tricarboxylate hydro-lyase. Enzyme Commission Number: EC 4.2.1.79. CAS No. 80891-26-5. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder. EXWM-5063; 2-methylcitrate dehydratase; EC 4.2.1.79; 80891-26-5; 2-methylcitrate hydro-lyase; PrpD; 2-hydroxybutane-1,2,3-tricarboxylate hydro-lyase. Cat No: EXWM-5063.
methanogen homoaconitase
This enzyme catalyses several reactions in the pathway of coenzyme-B biosynthesis in methanogenic archaea. Requires a [4Fe-4S] cluster for activity. In contrast to EC 4.2.1.36, homoaconitatehydratase, this enzyme can catalyse both the dehydration of (R)-homocitrate to form cis-homoaconitate and the subsequent hydration reaction that forms homoisocitrate. In addition to cis-homoaconitate, the enzyme can also catalyse the hydration of the physiological substrates dihomocitrate and trihomocitrate as well as the non-physiological substrate tetrahomocitrate. cis-Aconitate and threo-DL-isocitrate cannot act as substrates, and (S)-homocitrate and trans-homoaconitate act as inhibitors of the enzyme. Group: Enzymes. Synonyms: methanogen HACN. Enzyme Commission Number: EC 4.2.1.114. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder. EXWM-4954; methanogen homoaconitase; EC 4.2.1.114; methanogen HACN. Cat No: EXWM-4954.
Native Porcine Aconitase
Aconitase catalyses the stereo-specific isomerization of Citrate to Isocitrate via cis-aconitate in the tricarboxylic acid cycle. Aconitase is inhibited by cyanide, sulfide and copper and mercury at low concentRations. It is competitively inhibited by trans-aconitate. Aconitase contains an internal Fe-S cluster which is responsible for its activity. Crude preparation containing aconitase activity. Applications: Aconitase may be usefull in diabetes research as well as to study structural conservation between iron-responsive element binding proteins (ire-bp) and isomerases. Group: Enzymes. Synonyms: cis-aconitase; aconitase; Citrate hydro-lyase; Citrate (Isocitrate) hydro-lyase; EC 4.2.1.3; 9024-25-3; aconitatehydRatase. Enzyme Commission Number: EC 4.2.1.3. CAS No. 9024-25-3. Aconitase. Storage: -20°C. Source: Porcine heart. Species: Porcine. cis-aconitase; aconitase; Citrate hydro-lyase; Citrate (Isocitrate) hydro-lyase; EC 4.2.1.3; 9024-25-3; aconitatehydRatase. Cat No: NATE-0024.
phosphinomethylmalate isomerase
The enzyme, characterized from the bacterium Streptomyces viridochromogenes, is involved in bialaphos biosynthesis. The enzyme from the bacterium Kitasatospora phosalacinea participates in the biosynthesis of the related compound phosalacine. Both compounds contain the nonproteinogenic amino acid L-phosphinothricin that acts as a potent inhibitor of EC 6.3.1.2, glutamine synthetase. The similar enzyme EC 4.2.1.3, aconitatehydratase, cannot catalyse this reaction. Group: Enzymes. Synonyms: pmi (gene name). Enzyme Commission Number: EC 4.2.1.166. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder.
