Alanine Racemase (Crude Enzyme) This enzyme belongs to the family of isomerases, specifically those racemases and epimerases acting on amino acids and derivatives. This enzyme participates in alanine and aspartatemetabolism and D-alanine metabolism. It employs one cofactor, pyridoxal phosphate. At least two compounds, 3-Fluoro-D-alanine and D-Cycloserine are known to inhibit this enzyme. The D-alanine produced by alanine racemase is used for peptidoglycan biosynthesis. Peptidoglycan is found in the cell walls of all bacteria, including many which are harmful to humans. The enzyme is absent in higher eukaryotes but found everywhere in prokaryotes, making alanine racemase a great target for antimicrobial drug...udies have shown that without the alr gene being expressed, the bacteria would need an external source of D-alanine in order to survive. Therefore, the alr gene is a feasible target for antimicrobial drugs. This product with the indicated enzyme activity was briefly purified from engineered E. coli. Applications: Drug development; pharmacology; medicine; pharmacology. Group: Enzymes. Synonyms: L-alanine racemase. Enzyme Commission Number: EC 5.1.1.1. CAS No. 9024-06-0. Alanine Racemase. Activity: Undetermined. Appearance: Clear to translucent yellow solution. Storage: at -20 °C or lower, for at least 1 month. Source: E. coli. L-alanine racemase. Pack: 100ml. Cat No: NATE-1854. Mutant Alanine Racemase Y354N from Geobacillus stearothermophilus, Recombinant Mutant Alanine Racemase (Y354N) (mAR-Y354N), a pyridoxal 5-phosphate (PLP) dependent enzyme catalyzes the interconversion of the L-Serine to D-Serine. In WT Alanine Racemase Tyr354 plays a crucial role in defining the strict specificity of AR for alanine, in converting L-Alanine to D-Alanine, which is an important component of the peptidoglycan layer of bacterial cell wall. By mutating the active site Tyr 354 to Asn, the specificity of the enzyme changes and it becomes a racemase with dual specificity for L- Alanine and L-Serine. Applications: Mutant alanine racemase y354n can be used to convert l-serine to d-serine and l-alanine to d-alanine. Group: Enzymes. Synonyms: Alanine Racemase Y354N; alr; dal. Enzyme Commission Number: EC 5.1.1.1. Purity: > 99% by SDS-PAGE. Alanine Racemase. Mole weight: 43 kDa. Activity: > 50 mU/mg. Storage: Reconstituted enzyme can be stored in working aliquots at -20°C and use within 3 months. Avoid repeated freeze-thaw cycles. Form: Lyophilized. Source: E. coli and fused to His-tag at N-terminus. Species: Geobacillus stearothermophilus. Alanine Racemase Y354N; alr; dal; Alanine Racemase. Cat No: NATE-1639. Creative Enzymes
Native Bacillus stearothermophilus Alanine Racemase Alanine racemase is involved in alanine, aspartate and D-alanine metabolism. 3-Fluoro-D-alanine and D-Cycloserine are known to inhibit alanine racemase. Alanine racemase monomer is composed of two domains, an eight-stranded α/β barrel at the N-terminus and a C-terminal domain. The N-terminus includes residues 1-240, whereas the C-terminal comprises of the β-strand (residues 241-388). One molecule of pyridoxalphosphate (PLP) is present as the cofactor in each subunit. Applications: Alanine racemase is used to convert l-alanine into d-alanine. alanine racemase, from creative enzymes, has been used to isomerize l-[u-14c]alanine to a racemic mixture of l/d-[14c]alanine. Group: Enzymes. Synonyms: Alanine Racemase; EC 5.1.1.1; 9024-06-0; L-alanine racemase. Enzyme Commission Number: EC 5.1.1.1. CAS No. 9024-06-0. Alanine Racemase. Mole weight: Mr 78 kDa (2 subunits 39 kDa each). Activity: > 10 Iunits/mg solid. Storage: -20°C. Form: lyophilized powder. Lyophilized from 50 mM phosphate buffer, pH 7.5. Source: Bacillus stearothermophilus. Alanine Racemase; EC 5.1.1.1; 9024-06-0; L-alanine racemase. Cat No: NATE-0045. Creative Enzymes
aspartate racemase Also acts, at half the rate, on L-alanine. Group: Enzymes. Synonyms: D-aspartate racemase; McyF. Enzyme Commission Number: EC 5.1.1.13. CAS No. 37237-56-2. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder. EXWM-5371; aspartate racemase; EC 5.1.1.13; 37237-56-2; D-aspartate racemase; McyF. Cat No: EXWM-5371. Creative Enzymes
D-aspartate ligase This enzyme forms part of the peptidoglycan assembly pathway of Gram-positive bacteria grown in medium containing D-Asp. Normally, the side chains the acylate the 6-amino group of the L-lysine residue contain L-Ala-L-Ala but these amino acids are replaced by D-Asp when D-Asp is included in the medium. Hybrid chains containing L-Ala-D-Asp, L-Ala-L-Ala-D-Asp or D-Asp-L-Ala are not formed. The enzyme belongs in the ATP-grasp protein superfamily. The enzyme is highly specific for D-aspartate, as L-aspartate, D-glutamate, D-alanine, D-iso-asparagine and D-malic acid are not substrates. In Enterococcus faecium, the substrate D-aspartate is produced by EC 5.1.1.13, aspartate racemase. Group: Enzymes. Synonyms: Aslfm; UDP-MurNAc-pentapeptide:D-aspartate ligase; D-aspartic acid-activating enzyme. Enzyme Commission Number: EC 6.3.1.12. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder. EXWM-5715; D-aspartate ligase; EC 6.3.1.12; Aslfm; UDP-MurNAc-pentapeptide:D-aspartate ligase; D-aspartic acid-activating enzyme. Cat No: EXWM-5715. Creative Enzymes
