The enzyme from Streptomyces specifically hydrolyses the terminal lacto-N-biosyl residue (β-D-Gal-(1?3)-D-GlcNAc) from the non-reducing end of oligosaccharides with the structure β-D-Gal-(1?3)-β-D-GlcNAc-(1?3)-β-D-Gal-(1?R). Lacto-N-hexaose (β-D-Gal-(1?3)-β-D-GlcNAc-(1?3)-β-D-Gal-(1?3)-β-D-GlcNAc-(1?3)-β-D-Gal-(1?4)-D-Glc) is hydrolysed to form first lacto-N-tetraose plus lacto-N-biose, with the subsequent formation of lactose. Lacto-N-Biosidase, Recombinant
Lacto-N-Biosidase may be used during glycoprotein analysis to better study the structure and function of glycoprotein and glycolipid sugar chains. This product can be used for specific oligosaccharide hydrolysis of type-I chain oligosaccharides, producing lacto-N-biose (Gal β 1-3 GlcNAc) as a byproduct, but not oligosaccharide hydrolysis of type-II chain oligosaccharides. As a result, Lacto-N-Biosidase can be used to distinguish type-I versus type-II glycoprotein as well as glycolipid sugar chains. This enzyme is involved in the lacto-N-biose I/galacto-N-biosedegradation pathway in the probiotic bacterium Bifidobacterium longum. Differs from EC 2.7.1.157, N-acetylgalactosamine kinase, as it can phosphorylate both N-acetylgalactosamine and N-acetylglucosamine at similar rates. Also has some activity with N-acetyl-D-mannosamine, D-talose and D-mannose as substrate. ATP can be replaced by GTP or ITP but with decreased enzyme activity.
