MHY1485 markedly increased the LC3II/LC3I ratio dose-dependently and time-dependently by inhibition of the fusion between autophagosomes and lysosomes, and without increasing the autophagic flux. At 2 μM, MHY1485 did not show any cell death during longer treatment, supporting that MHY1485 had less toxicity than other well-known inhibitors of autophagy. MHY1485 induced cellular LC3-II accumulation in Ac2F is reported to be a result of autophagy inhibition due to reduced fusion between autophagosomes and lysosomes.
