Granzyme B from Mouse, Recombinant
Granzyme B, a 247 amino acid polypeptide, contains a leader sequence, which is cleaved by a signal peptidase and a two amino acid prodomain, which is cleaved by the lysosomal cysteine protease DPPI. The recombinant granzyme B is expressed in Pichia pastoris as the mature form and appears on SDS-PAGE as a triplet (~34, 32, and 30 kDa) due to three different glycosylations. Granzyme B, a serine protease, is the most prominent granzyme in a family of 11 found in the cytotoxic granules. The granzymes enter the target cell with the assistance of perforin, a critical molecule of the cytotoxic granules. In the target cell, the granzymes act on specific substrates involved with the cell death via apoptosis. > 90% (sds-page), recombinant, expressed in pichia pastoris, buffered aqueous solution. pichia expressed recombinant granzyme b has the advantage over native granzyme b as the latter can be purified only in limited amounts, and over e. coli and vaccinia virus expressed enzymes that failed to generate soluble active enzyme. Group: Enzymes. Synonyms: g. CAS No. 143180-74-9. Granzyme B. Mole weight: 28.9 kDa. Activity: > 10,000 units/mg protein. Storage: -70°C. Form: buffered aqueous solution. Source: Pichia pastoris. Granzyme K from Human, Recombinant
Granzyme K is a protein that in humans is encoded by the GZMK gene. This gene product is a member of a group of related serine proteases from the cytoplasmic granules of cytotoxic lymphocytes. Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein described here lacks consensus sequences for N-glycosylation present in other granzymes. Group: Enzymes. Synonyms: Granzyme K; GZMK; GZMK. Enzyme Commission Number: EC 3.4.21.-. caspase-3
Caspase-3 is an effector/executioner caspase, as are caspase-6 (EC 3.4.22.59) and caspase-7 (EC 3.4.22.60). These caspases are responsible for the proteolysis of the majority of cellular polypeptides [e.g. poly(ADP-ribose) polymerase (PARP)], which leads to the apoptotic phenotype. Procaspase-3 can be activated by caspase-1 (EC 3.4.22.36), caspase-8 (EC 3.4.22.61), caspase-9 (EC 3.4.22.62) and caspase-10 (EC 3.4.22.63) as well as by the serine protease granzyme B. Caspase-3 can activate procaspase-2 (EC 3.4.22.55). Activation occurs by inter-domain cleavage followed by removal of the N-terminal prodomain. Although Asp-Glu-(Val/Ile)-Asp is thought to be the preferred cleavage sequence, the enzyme can accommodate different residues at P2 and P3 of the substrate. Like caspase-2, a hydrophobic residue at P5 of caspase-3 leads to more efficient hydrolysis, e.g. (Val/Leu)-Asp-Val-Ala-Asp? is a better substrate than Asp-Val-Ala-Asp?. This is not the case for caspase-7. Belongs in peptidase family C14. Group: Enzymes. Synonyms: CPP32; apopain; yama protein. Enzyme Commission Number: EC 3.4.22.56. CAS No. 169592-56-7. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder. EXWM-4231; caspase-3; EC 3.4.22.56; 169592-56-7; CPP32; apopain; yama protein. Cat No: EXWM-4231.
Caspase 3 Human, Recombinant
Caspase 3 is a member of the CED-3 subfamily of caspases and is responsible for the cleavage of many key proteins such as the nuclear enzyme poly (ADP-ribose) polymerase (PARP), the inhibitor of caspase-activated deoxyribonuclease (ICAD), and gelsolin, a protein involved in apoptosis regulation. Caspase 3 is considered to be an effector caspase, activating pro-caspase 6 and pro-caspase 9 in vitro. Caspase 3 can be activated by caspase 8, caspase 6, and granzyme B. Human recombinant c-terminal histidine tagged caspase 3 is a fully active protein consisting of 17 kda and 13.5 kda subunits; the 13.5 kda subunit contains the histidine tag. Applications: Caspase-3 is a caspase prote...ve enzyme. this protein cleaves and activates caspases 6 and 7; and the protein itself is processed and activated by caspases 8, 9, and 10. it is the predominant caspase involved in the cleavage of amyloid-beta 4a precursor protein, which is associated with neuronal death in alzheimer's disease. alternative splicing of this gene results in two transcript variants that encode the same protein. caspase-3 shares many of the typical characteristics common to all currently-known caspases. for example, its active site contains a cysteine residue (cys-163) and histidine residue (his-121) that stabilize the peptide bond cleavage of a protein sequence to the carboxy-terminal side of an
Native Human Cathepsin G
Cathepsin G is an enzymatic protein belonging to the peptidase or protease families. In humans, it is coded by the CTSG gene. The protein encoded by this gene, a member of the peptidase S1 protein family, is found in azurophil granules of neutrophilic polymorphonuclear leukocytes. The encoded protease has a specificity similar to that of chymotrypsin C, but it is most closely related to other immune serine proteases, such as neutrophil elastase and the granzymes. Cathepsin G may participate in the killing and digestion of engulfed pathogens, and in connective tissue remodeling at sites of inflammation. It also localizes to Neutrophil extracellular traps (NETs), via its high affinity for DNA, an unusual property for serine proteases. Transcript variants utilizing alternative polyadenylation signals exist for this gene. Group: Enzymes. Synonyms: CTSG; cathepsin G; CG; CATG; EC 3.4.21.20; chymotrypsin-like proteinase; neutral proteinase. Enzyme Commission Number: EC 3.4.21.20. CAS No. 56645-49-9. Purity: > 96% (SDS-PAGE). CTSG.
