lipoyl synthase This enzyme is a member of the 'AdoMet radical' (radical SAM) family, all members of which produce the 5'-deoxyadenosin-5'-yl radical and methionine from AdoMet [i.e. S-adenosylmethionine, or S-(5'-deoxyadenosin-5'-yl)methionine], by the addition of an electron from an iron-sulfur centre. The radical is converted into 5'-deoxyadenosine when it abstracts a hydrogen atom from C-6 and C-8, leaving reactive radicals at these positions so that they can add sulfur, with inversion of configuration. This enzyme catalyses the final step in the de-novo biosynthesis of the lipoyl cofactor, with the other enzyme involved being EC 2.3.1.181, lipoyl(octanoyl) transferase. Lipoylation is essential ...pathway involves EC 2.7.7.63, lipoate-protein ligase, which can lipoylate apoproteins using exogenous lipoic acid (or its analogues). Group: Enzymes. Native Bovine Phenylethanolamine N-Methyl Transferase Phenylethanolamine N-methyltransferase (PNMT) is the enzyme which catalyzes the N-methylation of norepinephrine thereby resulting in the formation of epinephrine as shown below: Norepinephrine + S-Adenosyl methionine (SAM) -------> Epinephrine. Nicotinamide N-Methyltransferase from Human, Recombinant Nicotinamide N-Methyltransferase (NNMT) methylates nicotinamide and other pyridine containing compounds. NNMT transfers a methyl group from S-adenosylmethionine (SAM) to nitrogen N1 of nicotinamide to produce 1-methylnicotinamide (MNA) and S-adenosylhomocysteine. MNA is a messenger molecule that increases neurite branching, serves as an anti-thrombotic, and has anti-inflammatory properties. The role of NNMT overexpression in cancers may be to alter epigenetic methylation patterns in two ways: by lowering the intracellular concentration of SAM, required by methyltransferase enzymes, and by depleting the available NAD+ by transforming nicotinamide to MNA. Sirtuins use NAD+ as a substrate to alter protein acetylation and ribosylation, including histone targets. Group: Enzymes. Synonyms: NNMT. Enzyme Commission Number: E.C. 2.1.1.1. S-Adenosyl-L-Methionine S-adenosyl-l-methionine (SAMe) is a naturally occurring substance which is a major source of methyl groups in the brain. The efficacy of SAMe in treating depressive syndromes and disorders is superior to that of placebo and comparable to that of standard tricyclic antidepressants. Since SAMe is a naturally occurring compound with relatively few side effects, it is a potentially important treatment for depression. S-adenosylmethionine (SAMe) is a dietary supplement used in the management of osteoarthritis (OA) symptoms. Studies evaluating SAMe in the management of OA have been limited to Non Steroidal Anti-inflammatory Drugs (NSAIDs) for comparison. tryptophan 2-C-methyltransferase The enzyme, characterized from the bacterium Streptomyces laurentii, is involved in thiostrepton biosynthesis. It is a radical SAM enzyme that contains a [4Fe-4S] center and a cobalamin cofactor. The enzyme first transfers the methyl group from SAM to the bound cobalamin, followed by transfer from methylcobalamin to L-tryptophan, resulting in retention of the original methyl group configuration. The second transfer is likely to involve a CH3 radical species formed from methylcobalamin by the concerted action of a partially ligated radical SAM [4Fe-4S]2+/1+ center. Group: Enzymes. It is the first of three steps leading to the formation of siroheme from uroporphyrinogen III. The second step involves an NAD+-dependent dehydrogenation to form sirohydrochlorin from precorrin-2 (EC 1.3.1.76, precorrin-2 dehydrogenase) and the third step involves the chelation of Fe2+ to sirohydrochlorin to form siroheme (EC 4.99.1.4, sirohydrochlorin ferrochelatase). In Saccharomyces cerevisiae, the last two steps are carried out by a single bifunctional enzyme, Met8p. In some bacteria, steps 1-3 are catalysed by a single mult...(incorrect). spermidine synthase The enzymes from the plant Glycine max and from mammalia are highly specific for putrescine as the amine acceptor. The enzymes from the bacteria Escherichia coli and Thermotoga maritima prefer putrescine but are more tolerant towards other amine acceptors, such as spermidine and cadaverine. cf. EC 2.5.1.22 (spermine synthase) and EC 2.5.1.23 (sym-norspermidine synthase). Group: Enzymes. Synonyms: aminopropyltransferase; putrescine aminopropyltransferase; spermidine synthetase; SpeE; S-adenosylmethioninamine:putrescine 3-aminopropyltransferase. Enzyme Commission Number: EC 2.5.1.16. CAS No. 37277-82-0. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder. sym-norspermidine synthase The enzyme has been originally characterized from the protist Euglena gracilis. The enzyme from the archaeon Sulfolobus solfataricus can transfer the propylamine moiety from S-adenosyl 3-(methylthio)propylamine to putrescine, sym-norspermidine and spermidine with lower efficiency. cf. EC 2.5.1.16 (spermidine synthase) and EC 2.5.1.22 (spermine synthase). Group: Enzymes. Synonyms: S-adenosylmethioninamine:propane-1,3-diamine 3-aminopropyltransferase. Enzyme Commission Number: EC 2.5.1.23. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder.
